بازدارندگی سودوموناس‌های فلورسنت علیه گاایی نماتد Meloidogyne javanica روی گوجه‌فرنگی در شرایط گلخانه

چکیده :

از ریزوسفر بوته‌های گوجه‌فرنگی و درختان زیتون استان‌های گلستان، گیلان (منطقه رودبار) زنجان و قم نمونه‌های خاک جمع‌آوری شد. از این نمونه‌ها باکتری‌های جنس Pseudomonas روی محیط کشت King’s B جدا و ویژگی‌های فنوتیپی آن‌ها بررسی گردید. همه استرین‌ها روی محیط King’s B  رنگ فلورسنت تولید نموده و ویژگی‌های آرژنین دی‏هیدرولاز، رشد در چهار و 41 درجه سانتی‌گراد و اکسیداز در آن‌ها مثبت بود. بیشتر استرین‌ها نیترات را احیاء و ژلاتین را ذوب کردند ولی قادر به ایجاد واکنش فوق حساسیت در توتون، ‌تولید لوان روی محیط آگار غذایی حاوی 5% ساکاروز و فعالیت پکتولیتیکی روی حلقه‏های سیب‌زمینی نبودند. مایع فیلتر شده از کشت 48 ساعته استرین‌ها در محیط کشت مایع  King’s Bتهیه شد. اثر نماتدکشی این مایع روی لاروهای تازه تفریخ شده
Meloidogyne javanica در شرایط آزمایشگاه بررسی شد. نتایج نشان داد که در شرایط آزمایشگاه در مجموع بیش از 60% از استرین‌های مورد بررسی توانستند درصد قابل توجهی از نماتدها (بیش از 50%) را کنترل کنند. اثر نماینده استرین‌های با بازدارندگی بالا به همراه نماتدکش نماکور گرانول 10% روی تولید تخم، تعداد لارو و نماتد بالغ تولید شده، گاایی نماتد و وزن تر ریشه واندام‌های هوایی گیاه گوجه‌فرنگی در شرایط گلخانه بررسی شد. در شرایط گلخانه تعداد کل تخم، لارو، ماده بالغ و تعداد گال تشکیل شده روی ریشه‌های گوجه‌فرنگی تفاوت معنی‌دار نشان داد. بالاترین میزان تخم، لارو و ماده بالغ و تعداد گال تشکیل شده روی ریشه گیاهان شاهد بود. کمترین آنها در گیاهان تیمار شده با Pseudomonas fluorescens استرین 99 بود. بین تیمارها از نظر وزن تر ریشه و اندام‌های هوایی گوجه‌فرنگی اختلاف معنی‌داری وجود نداشت ولی با شاهد تفاوت نشان دادند. تیمار شاهد دارای بـالاترین میزان وزن تر ریشه و اندام هوایی بـود.

Inhibition of gall formation of Meloidogyne javanica on tomato using fluorescent pseudomonads under green-house condition

Abstract:

Soil samples were collected from rhizosphere of tomato and olive plants in Golestan, Gilan, Zanjan and Qom provinces (Iran). Fluorescent pseudomonas bacterial strains were isolated on King’s B medium and their phenotypic features were characterized. For all tested strains production of fluorescent pigments on King’s B medium, arginine dihydrolase, growth at four and 41 C and oxidase were positive. Most of them reduced nitrate and produced gelatinase but tobacco hypersensitivity reaction (HR), levan formation and pectinolytic activity on potato slices were negative for all. Filtrate liquid was obtained from 48 h culture of the fluorescent pseudomonas on King’s B broth medium. Nematicidal activity of the bacterial filtrate against fresh hatched Meloidogyn javanica juveniles was investigated under laboratory condition. Results indicated that above 60% of the tested strains showed nematicidal activity (>50% nematode mortality) under this condition. Efficacy of the representative of the most effective strains along with the nematicide, Nemacur 10G against nematode egg production, the juveniles, mature females and gall formation, plant fresh root and aboveground weight of tomato were studied under green-house condition. Under this condition total nematode eggs, juveniles, mature females and gall formation on tomato roots were decreased significantly as compared to other treatments.
 The highest number of eggs,, juveniles,, mature females and gall formation on tomato roots were observed in control treatment, and the lowest were recorded in plants treated with Pseudomonas fluoresces strain 99. There was no significant differences between the treatments as far as fresh root and aboveground weight are concerned, however, they differed significantly as compared to control check treatments in which the highest fresh root and aboveground weight.
